PLoS Biology | http://biology.plosjournals.org Of the 300 or so viruses that cause disease in humans, HIV may have the greatest adaptive advantage. Like most persistent viruses—including the herpesviruses Epstein–Barr and cytomegalovirus (CMV)—HIV employs various strategies to counteract its host’s response to infection. But HIV possesses a unique ability to sustain a progressive attack on the immune system—infecting the very cells that coordinate the immune response—leaving the body susceptible even to normally harmless microorganisms. It is these so-called opportunistic infections, rather than the human immunodefi ciency virus itself, that makes HIV so deadly. The specifi c mechanisms that engineer this ongoing systemic attack have been the subject of intense research. HIV targets white blood cells with protein surface receptors called CD4. These CD4, or helper, T-cells normally orchestrate the body’s immune response by signaling killer T-cells (which are also called CD8 T-cells, after their CD8 surface receptors) and other immune cells to multiply and differentiate—that is, become specially equipped to recognize a particular pathogen, or antigen. At the onset of infection, the immune system appears to respond normally, with a strong attack led by HIV-specifi c CD8 T-cells that initially contain the virus. But as the infection progresses, CD4 counts drop and the body’s ability to renew T-cells decreases while its proportion of “antigen-experienced’’ CD8 T-cells increases. While the biological effect of this hyperactivity is unclear, it is apparent that patients with elevated immune activity face a poor prognosis. Investigating the interaction among immune activation, CD8 T-cell differentiation, and HIV prognosis, Victor Appay and colleagues report that a close connection between elevated immune activation and elevated levels of highly differentiated T-cells may bring further insights into how HIV exhausts the immune system. To examine the effect of elevated immune activation on T-cells, the researchers analyzed T-cells from a group of HIV-infected individuals collected at two distinct points in time: at the onset of acute infection—which is characterized

Dynamics of Cortical Decision Circuits during Changes in the Fidelity of Sensory Representations

Every waking moment, we make decisions, from where to move our eyes to what to eat for dinner. The ease and speed with which we do this belie the complexity of the underlying neuronal processing. In the visual system, every scene is processed via a complicated network of neurons that extends from the retina through multiple areas in the visual cortex. Each decision requires rapid coordination o...

نورون های آینه ای و نقش آن درتقلید و نظریه ی ذهن در کودکان طیف اتیسم

Recent studies have reported the discovery of a group of neurons in the motor cortex of the brain's gray. These neurons are called mirror neurons. Rizzolatti and colleagues in 1990s discovered the first mirror neuron system in monkeys in the University of Parma. Mirror neurons in brain are responsible for understanding the actions and intentions of others. The mirror neuron system has an import...

Prefrontal delay-period activity reflects the decision process of a saccade direction during a free-choice ODR task.

To examine how delay-period activity participates in the decision of a saccade direction, we analyzed prefrontal activity while monkeys performed 2 tasks: oculomotor delayed-response (ODR) and self-selection ODR (S-ODR) tasks. In the ODR task, monkeys were required to make a memory-guided saccade to the cue location after a 3-s delay. In the S-ODR task, 4 identical visual cues were presented si...

Human amnesia and animal models of amnesia: performance of amnesic patients on tests designed for the monkey.

The performance of amnesic patients was assessed on five tasks, which have figured prominently in the development of animal models of human amnesia in the monkey. The amnesic patients were impaired on four of these tasks (delayed nonmatching to sample, object-reward association, 8-pair concurrent discrimination learning, and an object discrimination task), in correspondence with previous findin...

Dopamine neurons learn to encode the long-term value of multiple future rewards.

Midbrain dopamine neurons signal reward value, their prediction error, and the salience of events. If they play a critical role in achieving specific distant goals, long-term future rewards should also be encoded as suggested in reinforcement learning theories. Here, we address this experimentally untested issue. We recorded 185 dopamine neurons in three monkeys that performed a multistep choic...

How Laminar Frontal Cortex and Basal Ganglia Circuits Interact to Control Planned and Reactive Saccades Abbreviated Title: Frontal Cortex and Basal Ganglia Saccade Control

Acknowledgements J. Abstract How does the brain learn to balance between reactive and planned behaviors? The basal ganglia and frontal cortex together allow animals to learn planned behaviors that acquire rewards when prepotent reactive behaviors are insufficient. This paper proposes a new model, called TELOS, to explain how laminar circuitry of the frontal cortex, exemplified by the frontal ey...
